GLP-1 and GIP Receptor Biology
The incretin system
After eating, gut cells release hormones — including GLP-1 (from L-cells) and GIP (from K-cells) — that amplify insulin secretion in a glucose-dependent way. This 'incretin effect' is the foundation of a major research field.
GLP-1 receptors
Expressed in pancreatic islets, gut, and central nervous system. Established in research: glucose-dependent insulin secretion, glucagon suppression, slowed gastric emptying, central satiety signalling.
GIP receptors
Expressed in islets and adipose tissue among other sites. Established: insulinotropic action in experimental settings. Still studied: adipose lipid-handling effects and central actions — findings here remain partly investigational.
Why researchers combine them
Both receptors influence overlapping but non-identical pathways. Dual agonism tests whether combined engagement produces effects beyond either alone — the hypothesis behind tirzepatide and the rationale extended further by triple agonists.
