Origins
The incretin effect — greater insulin response to oral than intravenous glucose — was characterised decades ago and traced to gut hormones GLP-1 and GIP. That observation built an entire therapeutic and research area.
The modern landscape
Research now spans single agonists (GLP-1), dual agonists (GIP+GLP-1), triple agonists (+glucagon), amylin-analogue research (cagrilintide) and combination studies. Each generation tests whether broader pathway engagement changes outcomes.
Reading the literature critically
- Check the setting — in-vitro, animal, or human trial; each answers different questions.
- Check the phase — phase 1/2 signals are exploratory; phase 3 is confirmatory.
- Check the population — trial findings apply to the studied population, not universally.
- Separate mechanism from outcome — a plausible mechanism is not proof of effect.

Reference compounds let laboratories run analytical and in-vitro work — assay development, binding studies, method validation. They are tools for science, not consumer products.
